The first section of the table contains 37 markers, but some participants may have analyzed only the first 12 or 25. Each of the markers can exist as several variants, or alleles, which contain different numbers of a repeat unit, as the result of mutations from the original ancestor of all men. The figures below the DYS labels are the number of repeat units within each of that person’s markers or loci. This set of numbers for a given individual constitutes his Y-DNA haplotype. The second section of the table contains the results for markers 38-67, which several of the participants have had analyzed. The third section contains results for markers 68-111.
The makeup of the haplotypes provides useful information on two different subjects. (1) How closely related are two men? (2) Where was the original source of a man’s Lambert line? The first question is concerned with the recent genetic history of the individual and is most closely related to genealogy. The second question, expressed more technically, asks to what ancestral haplogroup does a man’s hyplotype belong. This question is concerned with older history having less genealogical content. It may provide considerable insight, however, into the ancient history of a man’s surname line.
RECENT GENETIC HISTORY
To compare two individuals, the DYS numbers are examined for each marker. If they have identical numbers at every marker (12 out of 12, 25/25, 37/37), the mismatch number is said to be zero. If 11 of the 12 (or 24 of the 25) are identical, then the mismatch number is one, and so on. In this way the mismatch number is calculated between two individuals. Different numbers of repeat units occur because of mutations, which can either insert another repeat unit or delete one, thereby either increasing or decreasing the number by one.
Geneticists have calculated how many generations it takes on average to create these differences. The probability of a mutation corresponds to a chance of about 0.002 (or 0.2%; 100% corresponds to a chance of 1.0) per marker per generation (one mutation every 500 generations per marker). To simplify the interpretation, only single mutations are considered at each marker, that is, either the markers agree or they do not. This assumption underestimates the number of mutations, because a difference of two repeat units between two individuals could result either from a single mutation that drops off two units in a single event or from two mutations that each drop off one unit.
A major objective is to establish how many generations have intervened between two living individuals and their most recent common ancestor. Consider the follow table taken from data on the FamilyTreeDNA website.

Two individuals with 12 matches and no mismatches have a 50% probability of having a common ancestor within 7 generations (210 years ago for a 30 year generation, or about the year 1800). This probability level is not very high, as it means the conclusion is wrong half the time. At the more acceptable 90% level, when the conclusion is wrong only a tenth of the time, it takes 23 generations to reach the common ancestor (about 700 years, beyond most genealogical proofs based on historical records). Thus even a perfect match with the 12 marker test provides only a crude indication of a close relationship between two individuals, although it is perfectly able to tell when two individuals have little relationship. Negative conclusions are more easily achieved in this business (“two people are NOT related”). Positive conclusions (“two people ARE related”) are harder to achieve. If two individuals have 11 matches and just one mismatch, the 50% probability requires a nearly useless 17 generations (about 500 years) to reach the common ancestor. These people are only possibly related. With ten matches and two mismatches, it is unlikely the two individuals are related, as the distance to the common ancestor moves into the thousands of years.
Things look less bleak with the 25 marker test. Two individuals with 25 matches and no mismatches have a 50% probability of a common ancestor within just 3 generations (90 years, or about 1910), a very reasonable target for many paper genealogies. With 90% probability, the common ancestor is 10 generations back (300 years or about 1700), still a reasonable target. With 24 matches and one mismatch, the 50% probability is 7 generations back (210 years or about 1800), although at the 90% level 16 generations (ca. 1500) are required. Two individuals with these results definitely are related. Even with 23 matches and two mismatches, 50% probability requires 11 generations to the closest common ancestor. The more reliable 90% probability, however, requires 23 generations (ca. 1300). Such men are probably related. Because mutation rates can vary widely at different loci, these numbers are not general but provide general guidelines.
With the 37 marker test, the results become even more reliable. Matches of 37/37 or 36/37 imply very closely related individuals, and matches of even 35/37 and 34/37 indicate closely related men. Only with six or more mismatches are the two men definitely unrelated. With these numbers in mind, we now can turn to the actual results with Lambert men.
The results of the analyses in the table at the top of this page now can be examined in light of this information. Nineteen individuals have been classified in Group A, as descendants of John Lambert Sr. of Rowan Co., NC, and Wilkes Co., GA (see The Lambert Surname). Many of these individuals had traced their ancestry back to this man, and others had discovered their descent only through this surname project. All but one (N32778) have a perfect 12/12 match. Normally, a difference of a single marker out of the first 12 would not be strong evidence for a recent common ancestor. In this case a single mutation occurred at DYS 385b at some point between John Lambert Sr. and the participating individual. John Lambert Sr. had at least seven sons, and descendants of six are represented in the project (James, John, William, George, Elijah, and Elisha, but not Thomas who likely had no male descendants). Interestingly, there are few non-Lambert exact 12/12 matches and no exact 25/25 matches in the entire FamilyTreeDNA database to this haplotype, indicating that it is relatively unusual. Fifteem of the 16 Group A participants have tests with 25 markers or more, and eleven of these individuals have a perfect 25/25 match. The other four have either 24/25 (N32778 and 10298) or 23/25 (9900 and 44512) matches. Thus the family begins to collect mutations. Note that the participant who had an 11/12 match now is at the 24/25 level, establishing his close relationship to the other members of Group A. The perfect 25/25 matches confirm the original haplotype (H1) for John Lambert Sr. and demonstrate that it is very stable. Thirteen of these individuals have obtained 37 marker tests, and more variability appears at this level. Eight individuals have a 37/37 match (6452, 7905, 10176, 10239, 10929, 63063, 96943, 121037) from three different sons (James, George, and Elisha). Their haplotype therefore represents that of John Lambert Sr. None of the remaining five individuals with 37 marker tests differs from the ancestral haplotype by more than three mutations, consistent with the distance from the present to John Lambert Sr. of about 7-10 generations. As yet, analysis of the mutations has not shed light on when specific mutations occurred. Mutations have been found in the lines of the sons John, William, and Elijah.
One certainty that emerged from these results is that the James Lambert who left a will in Wilkinson Co., GA, naming his wife Esther (usually transcribed Easter) is of the line of John Lambert Sr. of Wilkes Co., GA, and Onslow Co., NC (Group A). His descendants proved to have the haplotype of John Sr. There had been some question as to whether he was from this line or that of the Burke Co., GA, Lamberts (Group B). Clearly he is on the John Sr. line, something that could not have been proved by traditional genealogy. Three brothers (Blakely, Noah, and Lovinski/Lovzinsky) were associated with a Burke Co. group of Lamberts and hence were thought to be on the Group B line. Three participants descended from these brothers, however, have proved to be from Group A with haplotype H1. Their exact connection with John Lambert Sr. has not yet been established.
One of the questions unanswered by examination of historical records concerned whether the two groups of Lamberts in pre-1800 Georgia were related. The descendants of John Lambert Sr. (Group A) came from NC and moved into Wilkes Co., Ga, during the 1780s. The descendants of James Lambert (Revolutionary soldier) (Group B) moved into Burke Co., GA, about the same time from sources unknown. There are four participants descending from the Burke Co. group. They have identical 12-marker haplotypes. Together, they define the genetic fingerprint of the Burke Co. Lamberts as a stable haplotype. Two of these individuals have tested at the 25 marker level, and they have a 24/25 match. They differ only at DYS 464b. Thus at this stage we do not know the exact 25-marker haplotype of the Burke Co. ancestor, James Lambert. The Group B participants have 12/12 and 23/25 matches with individuals possessing surnames other than Lambert. More Group B individuals need to be tested to understand the group more fully, and 37 marker analysis may be useful. This group differs at seven sites from the 12-marker haplotype of the descendants of John Lambert Sr. of Wilkes Co., GA (Group A). Clearly, these two groups have no close relationship despite living in close proxmity in Georgia. Any common ancestor would have lived thousands of years ago.
Group C as defined traces its ancestry to Samuel Lambeth or Lambert of Craven Co., NC, in the late 1700s. Geographically, the ancestors of Group C were very close to Group A before the latter group moved from NC to GA. Participant 74199 presently is the only proved descendant of Samuel Lambeth in this project. Participant 17886, whose ancestors include men named Lambeth, has an exact 25/25 match with 74199. This individual’s line, however, has not been traced earlier than 1867 for certain, with residences in IL and AR. These two participants have a 90% probability of sharing a common ancestor within 10 generations, which corresponds closely with the expectation for living descendants of Samuel Lambeth. Consequently, we have placed him in Group C. Participant 96304 is descended from William Lamberth, who was born in SC in 1806. His father Frederick Lamberth was born in England. Thus there is no known connection with the Craven Co., NC, Lamberts/Lambeths, but his DNA places him in Group C. Participant 96363 descends from Robert Lamberth, who probably is a grandson of this same Frederick Lamberth. The line of particiapnt 97214 is known only as far as his grandfather, so his match with the others sheds no further light on their common lineage. All five members of Group C match 12/12, and the three with 25 markers match 25/25. They may be connected in England in the time period circa 1700 (10 generations with 90% confidence according to the above table). There are numerous matches at the 12 marker level by the Group C haplotype with men with other surnames in the FamilyTreeDNA database, so this haplotype is common. There is only one other non-Lambe(r)t(h) person with a 25/25 match, but many at the 24/25 or 23/25 level. Individuals in Group C require at least 25 markers to distinguish them from men with other surnames, and 37 markers would be extremely useful. These five individuals differ from the Group A haplotype by two markers out of 12 or four markers out of 25. These differences suggest some 60 generations (nearly 2000 years) to the common ancestor, at the 50% confidence level. Although this distance is large, Group C still includes the closest relatives to Group A currently in the Lambert surname project.
Individuals 14731 and 224401 are descendants of Joel Lambert of Henderson Co., KY, who in turn is a descendant of the Mecklenburg Co., VA, Lamberts. Together, they define the haplotype for Group D. The haplotype of 14731 differs from that of Group A by 10 out of 12 markers and from that of Group B by 7 out of 12. Thus all three southern Lambert groups come from very different genetic sources, with common ancestors probably several thousand years ago.
Individual 21048 originally thought he was a descendant of John Lambert Sr. of Wilkes Co. (Group A) through his son John Jr. His haplotype, however, differs from that of Group A by four markers out of 12. Such a difference requires a common ancestor several thousand years ago and eliminates the possibility that this person belongs to Group A. This person’s next recent ancestor is Richard Lambert (ca. 1770-ca. 1860). There is no paper record that John Jr. had a son of this name. The next most recent ancestor (Richard’s son) is Hugh Lambert, a name found in Group D. Individual 21048, however, differs from the haplotype of 14731 by nine markers out of 12. There appears, therefore, to be no connection between 21048 and Group D. This person has an exact 12/12 fit with individual 51783, whose ancestor lived in SC, and both of these people match 39466 at the 11/12 level. Both the first two individuals, however, also match over 20 non-Lambert individuals at the 12/12 level. Apparently they have a relatively common haplotypye. To determine how closely the three men are related would require 25 or 37 marker tests. At present only one of the three has gone beyond the 12 marker test.
Three individuals (N16959, 70265, and 104375) have 25/25 matches and are designated Group E. Two other individuals (N50253 and 107038) match 24/25 with these three. A sixth individual (119360) at present as a 12/12 match with the other five. Three of these five have 37/37 matches, and the other two match 36/37. Two even match 67/67. Individuals N50253, 70265, and 107038 descend from Peter Lambert of Lee Co., VA (late 1700s), son of Johann Heinrich Lambert of Wyeth Co., Va, (d. 1814) and of Otterberg, Palatinate (Pfalz), Germany (b. 1736). Individual N16959 is probably from this same line. Individual 104375 descends from Peter’s brother Henry (b. 1775). Individual 119360 probably descends from a third brother, John. Participant 70265 originally was thought to have a line of descent from George Edward Lambert of Kent Co., MD (late 1800s). The DNA results brought about a reassessment and then a reassignment. This group appears to be one of German ancestry that entered the American South at about the time of the Revolutionary War.
Three individuals (21048, 51783, and 102935) match at the 12 marker level and are designated Group F. It is possible that all three descend from a single individual, John Lambert of Yorkshire, England, b. ca. 1720. Individual 21048 is from this man’s son, Richard, who immigrated to NC or SC. The earliest ancestor John of individual 51783 may be a grandson of John of Yorkshire. The line of 102935 is out of GA, and the earliest ancestor James, born ca. 1790, also could be a grandson of John of Yorkshire.
There are three other pairs of individuals with 12/12 matches: 21184/73390, 38946/42242, and 62455/96038. In some such cases, known relatives were testing, but in others the identity was entirely a coincidence. Until further members are added to these pairs, we do not consider them an official subgrouping. The remaining participants possess unique haplotypes with no close relatives among the other participants.
DISTANT GENETIC HISTORY
Extensive genetic tests have been carried out on indigenous people throughout the world. These include individuals still living in the place where their very distant ancestors lived. Comparisons of their Y DNA analyses have generated a number of human groupings or haplogroups (see DNA Analysis For Genealogy).
The members of Group A, originally from Rowan Co., NC, and Wilkes Co., GA, all belong to the haplogroup I1. This group is associated with both Scandinavian and Anglo-Saxon populations. From the middle of the fifth century AD, the Angles, Saxons, and Jutes from Germanic areas on the Continent displaced the original British populations, which today include Scottish, Welsh, Cornish, and Irish in the west and north of Great Britain, as well as Ireland. Scandinavian (Viking) incursions into England and Ireland prior to the time of William the Conqueror were extensive, primarily from Denmark. The Normans, who began arriving in 1066, also were of Scandinavian stock (“Norsh Men”). The genetic results suggest that this Lambert line (Group A) came from one of these populations. There is a slightly higher likelihood for an Anglo-Saxon origin, as Group A differs from that of the prototype I1-AS (AS for Anglo-Saxon) at only four markers but from that of the prototype I1-N (N for Norse) at five. For example, at marker 390, the allele number 22 is common within the Anglo-Saxon group but 23 is common within the Norse or Scandinavian group. Members of Groups A, C, and F and two unaffiliated participants bear allele numger 22 at this site, consistent with an Anglo-Saxon rather than a Scandinavian course. Two unaffiliated participants bear allele number 23 at 390 and descended most likely from a Scandinavian source. Whereas I1-N peaks in Sweden, I1-AS peaks in the lowland Germanic areas of north Germany, Denmark, the Netherlands, the Norman areas of France, and parts of Great Britian.
Six individuals, without any known relationship, belong to haplogroup I2a. In contrast with I1, this haplogroup is not common in Scandinavia or western Europe. It is thought to have originated in eastern Europe and the Balkans. One explanation for people of English origin with this haplogroup is that their ancestors were Roman soliders, who commonly were from the eastern part of the Empire. Further testing could reveal to which subclades these individuals belong, such as I2a*, I2a1, or I2a2, some of which have associations with western Europe.
A single participant belongs to haplogroup I2b. Its distribution is similar to that of I1, but it is concentrated in the northern Germanic regions rather than in Scandinavia.
The four members of Group B from Burke Co., GA, and one unaffiliated participatnt belong to haplogroup R1a, as does individual 12450. This haplogroup is thought to have originated near the north side of the Black Sea and moved both west to eastern Europe and east to India and Eastern Asia as part of the original Indo-European culture. There are high concentrations (>50%) in Poland, the Ukraine, Hungary, and Kyrgyzstan, but still significant amounts (>20%) in Croatia, Russia, Norway, Iceland, and the Orkneys. Clearly, the roots of Groups A and B have no common ground for millennia, until they arrived in Georgia. Individual 12450 differs from Group B at four out of 12 and seven out of 25 sites, so there is no close relationship.
The remaining participants, including the sole member of Group D, belong to various subcategories of the large R1b haplogroup, particularly R1b1 and R1b1b2. This haplogroup family is not only the commonest among Lambert men but also the commonest in Europe. It includes the Atlantic modal haplotype and is thought to have originated with the men who repopulated Europe some 10,000 years ago after the most recent Ice Age. The Aurignacian culture from ca. 30,000 years ago has been associated with this group. Most Celtic populations have high proportions of this haplogroup. The original occupants of the British isles, including the Irish, Scottish, Welsh, and Cornish, were perdominantly R1b.
Lamberts have a wide variety of genetic antecedents. The surname clearly was assumed by numerous groups from many geographical and genetic sources. We have already identified Saxon/Scandinavian/Viking/Norman (haplotype I1), Balkan/East European (I2a), early Indo-European (R1a), and Celtic/British (R1b).
